I am vaccinating goats with a purfied protein + adjuvant. The animals generate high levels of IgG antibodies in their serum; however, I am interested in collecting milk from these animals and testing for IgA antibodies to my protein. I would also like to collect milk from the goats and purify/concentrate IgA antibodies as well. I am having a difficult time finding a procedure or reference that specifically addresses isolation of goat IgA.
Dear Thomas , i isolated many proteins from goat milk . the first step is to remove fat from milk by using centrifuge (7000 rpm /min at 4 C ). the second step is to remove of casein from milk by precipitation at pH 4.3 . After that you can separate IgA using ammonium sulfate and chromatography .
Dear Jasim, a couple of questions:
1. could you give me the g-forces required for centrifugation? We may have different rotors/centrifuges so we need to calculate speed based on x G forces.
2. what type of column do you use for separation of goat IgA?
3. Does this separate IgA with secretory factor?
4. do you have a protocol which could help?

Is there something like binding to a protein A or protein G column? I don't know if they work with goat IgA.
