If so, how many hours could I incubate the primary antibody? and in which temp (4 degrees or room temp ?

the section will be incubated preferably overnight under room temperature but in the morning you transfer the tissue section to microwave for 45 minutes so as to re-activate the antigens and protein in the tissue. the secondary antibody is usually for just 1 hour. i hope you find the response useful
