I've been doing PCR reactions using DNA extracted from a vaccine which contains aluminum hydroxide as adjuvant. I've tried with different conditions and quantities of template and DNA polymerase and I don't get results. The primers are fine, they have been tested with other samples different from vaccine
Any idea of how can I solve this problem?
Have you tried chelating the aluminum out of your preparation and then adding magnesium for your polymerase?
Nope, i havent tried that yet. I was just guesing if that was the possible reason i don't get any amplification, but it's a good advice, i'll try that. Thankyou Maria.
Do you dilute extracted DNA before performing the PCR? I used to have problems with PCR extracted from pear tree leaves, but the solution in my case is as simple as diluting 1/10 or 1/20 my samples in ultrapure water prior the PCR.
