Hi,

Why do you use a pH7 mobile phase? Is it for a post-column enzymatic reaction?

At this pH, microorganisms can pill in the mobile phase and clog the precolumn and the column, which results in an increase in pressure. Try to re-filter the mobile phase on a millipore filter.

In addition, the silica columns are not stable at high pH. Good luck.
