I have water samples from a river that were taken c.a. 1.5 years ago and were placed in the fridge, and now I am wondering if it pays the effort to proceed with a protocol to check the structure of the diatoms using microscopy. I have absolutely not experience with diatoms... Thanks!

Yes you can find the diatoms in that sample, because diatom cell wall is made up of silica and it will not degrade thousand of years. but for proper microscopy u have to follow some protocol.

thank you so much for the answers. I gues I will give it a try, also for the fun to do something new. @Joaquin, I did not fixed anything. What I have are c.a. 200ml of river waters in the fridge, without anything added. And my goal is basically to check the prescence of diatoms, which should be there because we saw them by 16S rRNA sequencing (chloroplasts).
