Hello, I am currently a master's student conducting research.
In my experiment, I deliver nanoparticles loaded sonosensitizers to cancer cells, and then apply high-frequency treatment to induce cancer cell death. According to various published studies, cells attached to well plates are typically treated at intensities above 1.0 W/cm² for more than 2 minutes. However, with our equipment, we observe that even at 0.5 W/cm² for over 2 minutes, the cells detach and die.
I would greatly appreciate any advice or suggestions on how to resolve this issue.
The cell line we are using is MCF-7.

MCF-7 is usually a well-adherent line. But, your experimental condition may have contributed towards that fragility that you observed. May I ask what passage number your MCF-7 cells are at?
After you observed this issue, did you try to do proper control to isolate the causes? You can run a control cells with ultrasound only to test whether your cells survive the treatment setup without the sonosensitizer or nanoparticles. If you're seeing significant cell death, then you'll need to optimize your ultrasound parameters before interpreting effects from your actual formulation.
Also, have you tried coating the wells with ECM proteins (e.g collagen, fibronectin, or gelatin)? It might gave better support for your cells to adhere during the treatment.
