Hello everyone. I was wondering if you can help me clarify this question. I am seeking to perform infection assays with bacteria, using this human cell line. Research articles regarding infection assays with bacteria and KG-1a are scarce.
They are already differentiated into macrophages but is there the need of using for instance PMA or some other compound to induce some response in these cells upon infection?

What makes you believe KG1a is a differentiated macrophage? I would view it as a transformed myeolid progenitor that is resistant to differentiation. I think you want to use THP1 or U937 for your experiment. (perhaps even primary macrophages...)

I am aware that THP-1 monocytes are also a good option for my experiments. Nevertheless KG-1a are the cells that I currently have in the lab and I wanted to test them, that is why I make the question.

KG1a is an AML human cell line and cannot be considered a "macrophage" cell line.There are reports of cytokine/phorbol esters induced differentiation of the parent KG1 cell line into macrophage-like cells by morphology, but a quick search reveals that the KG1a daughter cell line seems to be resistant to differentiation and retains a more primitive phenotype. I suggest you try to obtain the KG1 line and, maybe in the near future, try to work with BM or PB monocyte-derived macrophages for your experiments.
