benzoquinone. So I tried to oxidize 4-mercaptophenol (MP) using tyrosinase (TYR). Unfortunately, no matter how I change the TYR/MP concentration ratio, or incubation time, or incubation temperature, or pH, the reaction seems to be unsuccessful. In the last experiment, I chose 1mM PH=6.5 PBS buffer and room temperature. The final concentrations of TYR and MP are 1kU/L, 10kU/L and 0.1mM, respectively.And the incubation time is 1h. UV-vis results show almost no change. Any leads to resolve these issues are welcomed. Thank you in advance

Your spectra definitely show the differences among three spectra. Could you please normalise each spectrum and plot three spectra on the same axis.
Thank you for your response. i am sorry for that i could not understand what u mean. As the figures shown, it seems no obvious change even though I changed the concentration of TYR. And the two Figures all revealed that the spectra of the blank group（MP） almost overlaps with the experimental group（MP+TYR）. Besides that, i have checked that the UV characteristic peak of benzoquinone is around 400nm. In the two figures, no peaks show at 300-400nm.

What is the purpose for? Whether the spectrophotometer has a wavelength calibration issue if you think the materials should have the absorption between 300nm and 400 nm?
Whether you could use alternative material to Oxidation of 4-mercaptophenol ?
