Do I need to make a heat shock for my PCr-tubes to ensure DNA unwanding ?

I did three PCR for the same DNA sample. The first and second one gave me a clear band band. However, in the last PCR I put the tubes in the PCR machine before it starts the heating cycle (25 °C). To clarify, I did not make heat shock. Unfortunately, it did not give me a band in the agarose electrophoresis. Does it can be a reason.
