I'm working with H9C2 cell line from rat heart myoblast and I would like to investigate a bit more their electrophysiological properties after differentiation. Do they fire action potential spontaneously, like cardiomyocytes? Should we stimulate them in order to obtain any kind of electrical response? I'm curious about their electrical properties and would be happy to know more about it.

Dear Benoit: I do not think that H9c2 cells can fire action potentials, even they are well-differentiated. Once differentiated, these cells can exhibit L-type calcium current of skeletal or cardiac type. However, the magnitude of delayed rectifier K(+) currents, at particular, Kv2.1-encoded currents, appear to be so large to prevent generation of action potential.

H9C2 cell do not fire action potential, and indeed they do not show beating activity.
This being said, if you are looking for a cardiac cell line that fires action potentials spontaneously you may want to look at HL-1 cells https://www.ncbi.nlm.nih.gov/pmc/articles/PMC19680/
