I am using an ATP assay to detect cell viability. Do ATP assays only detect intracellular ATP?
My reasoning is that, due to ATPase, most extracellular ATP should be gone, so we should be measuring intracellular ATP in the assay.
Can someone let me know if I am correct?
There is no research to answer this question for sure but in general, I think everyone will agree with your reasoning. It is generally accepted that we are measuring intracellular ATP with ATP assay, so you should go ahead and not worry about it.
The best way to be sure there is no extracellular ATP being counted as intracellular ATP is to wash the cells to remove any that might be in the medium. You could also test the medium itself for ATP using the same assay. Then you could be sure.
