We need your experience with DNA concentration and purity measurements on UV5Nano Mettler Toledo spectrophotometer. We have problems in repeatability between measurements of same samples. We are using 3 microliters of extracted dsDNA from QiAcube Connect device by DNA Blood Mini kit (Qiagen, Germany).
We are using this data about DNA concentration for HLA typing and genotyping of blood groups on FluoQube inno-train's machine.
What is your suggestion for improving the measurement?

Why Aren't My NanoDrop Results Reproducible? and the common problems are a dirty optical path, sample and standard/blank are not jn exactly the same solvent and evaporation and gas absorption which can change with time so check if your blank is changing absorbance between replicates
Dear Paul, thank you for your answer. Unfortunately, we are using only very small content, only 3 microliters without cuvette. Nothing could be dirty, we are using the same solution in blank as the sample is eluted. Could be something else the reason? I am medical biochemist and have experience with spectrophotometry, but not with UV until now. Best regards, Jasna
