Dear all,

I am growing human primary airway epithelial cells from excised normal lung tissue in 2D ALI culture.

In order to validate my model, I am doing immunofluorescence staining of the various epithelial cell types.

I'm using SCGB1A1 as my marker for club cells (antibody cat#07-623, Sigma).

I've found that the staining tends to occur in clusters, with a granular type pattern on the apical surface of my membranes. This fits with a secreted type pattern, however I was wondering if other people are also noticing staining in clusters.

I've found two papers on the distribution of club cells, however they are more concerned with distribution throughout the airways from proximal to distal, as opposed to patterns.

What have other people observed?
