I am having trouble calculating the original concentration of my viral stock. I use 400 µl of my stock to isolate viral DNA, then eluate this in 30 µl of wich I add 3 µl to my qPCR. I also have a dilution curve as my standard curve (7 samples of known titer). With linear regression I can calculate my stock titer, I am just having touble incorporating the step of the DNA isolation as the dilution factor.

What I have come up with is x30/400*1000. The *1000 is due to me wanting to have my viral titer in particles / ml.

If anyone can advise on whether I have missed a step it would be greatly appreciated.
