Hello fellow researchers,
I am currently working on isolating Ophiognomonia leptostyla from acervuli on walnut leaves but have been facing issues with conidial germination on PDA plates. After 10 days, there has been no sign of germination.
Could anyone recommend the most suitable media and optimal conditions for isolating O. leptostyla? Additionally, are there any specific solutions or treatments that could help improve conidial germination?
Any suggestions or advice would be greatly appreciated!

I think to use gibberellins with mediam or pretreatment with it .but which method you used for germination?
First, determine if the seeds are viable using the flotation test. Seeds that sink in water are viable. Then only use these in your experiment :)
