Hi, I am attempting to prepare collagen hydrogels using Corning® Collagen I, Rat Tail (high concentration), which is supplied in liquid form in 0.02 N acetic acid. My objective is to create a stimuli-responsive hydrogel, and I am following standard protocols, including adding PBSx10, NaOH, DDW, and finally, the collagen. The collagen concentration in the hydrogels I've tested is at least 2mg/ml, with some trials using higher concentrations (3 and 4mg/ml). However, I am encountering two problems:

After the gel forms (verified with the inverse tube test), I add 300ul of PBSx1 to the vial. The gel floats, and the PBSx1 solution ends up beneath the gel.

I aim to conduct rheology analysis, but when I transfer the gel out of the vial, it releases a significant amount of water. This makes it challenging to perform measurements because the gel flows over the area when the upper plate begins to touch it. Note that the vials used are HPLC-grade glass vials.

I would greatly appreciate any help and information regarding the creation of hydrogels based on collagen.
