Yes Robert, this is my supposition. But since it's the first time that I have this kind of problem in peptide synthesis, I'd like to know if is this particular resin that gives this problems.
Hi Veronica, In DMF the resin swells and clog your filters. Try to find solvents that shrink the resin (non-solvent for the polymer). Then make a series of solvent mixtures from 100% DMF to 10% DMF in the non-solvent. Use this for washing. Always add small lots of solvents for washing instead of using entire solvent at a time. This will help. Sorry I do not remember non-solvent for SASRIn resin but you can find one. All the best.
I have worked with SASRIN resin, its time waste process. and one way, if you are using DCM means some fast removal is possible and why you are waiting for that much time to filter. you use down by vacuum means it will be fast too.
but processed air you need to allow up and down by vacuum means your problem will get resolve.,
I Agree with selva., its just like gel so this difficulty to remove solvent. so you try to give vacuum with processed air pressure means it will get remove.,
