I am looking into doing a peptide conjugation and the protocol im looking at says to use dialysis tubing to separate the reacted peptide from unreacted. I do not have much of the peptide so i wanted to reduce the reaction volume and use a protein concentrator for clean up.
I was wondering if dialysis tubing with a 6-8 kD MWCO would work the same as a protein concetrator with a 6-8kDa MWCO in my situation.
yes it could be used by tying them on MCT and keep them inverted in PEG
Surely you can use, the movement of unreacted peptide shall depend on it's molecular mass. It shall be smaller than cut off limit of dialysis member.
Alternatively, use a Sephadex G-25 or G-50 column to separate reacted and unreacted peptide to separate such molecules on the basis of individual molecular masses.
Thanks for the tips everyone. I like your suggestion Sebastian, seems like the simplest solution, not sure why I didnt think of it.
Additionally can anyone recommend an SEC column that would be able to able to resolve a 2kD and 2.5kD peak from each other? Im not sure if that is feasible with normal methods.
