I have been growing mouse organoids and they grow well at first. I am using the general media and conditions suggested by Clevers´lab and other labs. Before I provide more in detail information I would like first to share the picture of how the organoids look like after the first week, when they lose the cystic morphology and acquire this blob-like morphology, every day more cells acquire these big blobs and stop growing normally (medium does not turn yellow-ish). Have anyone experience this issue or have observed this phenotype before?If so, any suggestions about what may be causing it?
I am experiencing it now for mouse liver organoids. I am also searching for solution. If you know now, please share.
However, when I ask about this to other researchers, they suggested me not to use the same culture medium for more than 2 months as they lose the potential. And change the medium for every 2 to 3 days.
I use the mouse liver organoid normal culture medium from Organoid sciences. Their medium usually doesn't change the color to yellow even if the organoids grows well.
Can you please expand more on your comment? Could you specify which are the components of your medium and the concentrations?
They didn't reveal the components of the medium. However, the organoids are growing well for me now. As the "Organoid sciences" company stopped selling the culture medium, I am now making the culture medium by myself following this nature protocol.
