I add phenol red in my antibody so the waste will be red. Interestingly, every time after I strip my membrane with pH 2.3 stripping buffer and pour the waste into the container the yellow color is always at the bottom. I have seen this for 10 years. Does this mean acidic solution is heavier than alkaline solution?
Acidity is the quantity of H+in a solution. Basicity measures how much they lack those H+.pH7 is distilled water. If you try salt water it is around 8.If you try the cytosol, it is about 5-6.I supose dH20 will be acidic. Can it be an emulsion? Like a detergent? Red is 1-3.yellow is about 4-5.Membrane? Does this include protein?

I will caution that salt water pH is around 8. My nephrology professor even made a big mistake to put it into his textbook. In fact, most distilled water and saline (NaCl 0.9%) will be pH ~5.8 (his explanation was the Cl- is acidic) if you let them sit long enough (even in the sterilized container) due to CO2. If you bubble the solution with inert gas for a few seconds then they will go back to pH 7.0. My question may not have any scientific value but it is fun. Initially, I observed the phenomenon when I toss my phenol red containing antibody into the waste container then add acidic stripping buffer (not on purpose, but either albumin or skim milk was in it). When I added the waste in opposite sequence it gave me the same picture. I thought about protein might did the trick so I tried simple solution without protein which showed me the same thing. I am not a chemist but just have fun to see it.
The CO2 changes the pH. This is why it flips back. The CO2 makes the pH change.(to alkaline)?. I'm glad is fun. A detergent might desintegrate the membrane
I do not think Tween-20 will disintegrate membrane within a few minutes, otherwise we are all in big trouble.
