I have a question on plant disease sampling methods. I plan to collect wilt samples from different farms. I am rather confuse with the term, "isolates". Does 1 fungi isolate represent 1 farm in general? Or, does 1 fungi isolate represent 1 sample tree in that particular farm?
For example, if I collected 3 diseased samples from a farm and I have successfully isolated the pathogen, does that mean I have 3 isolates from that particular farm? If I have visited 3 different farms and similarly, I collected 3 diseased samples from each farm, does that mean I have 3 farms x 3 diseased samples = 9 isolates?

As per of my understanding, I think an isolate represent one particular geographical location.
Hi Wong That nice question, every pathogen isolated from each plant is call isolate, in the same farm. So you can have 10 isolate of pathogen from the same farm.
The sampling strategy you described is correct, each universe (in this case the farms) is an isolated. Now you have nine isolates. At the conclusion of the study you have isolates that correlate a geographical region or locality.

Isolate is a sample of pathogen that collected from a host and another isolate is a sample from other host but the host same the first one or a different host, it is depeneds on your work
Any microorganism you isolate from any host under specific environment is an isolate. that means, each plant is a different environment, so each sample you take is an isolate.
The definition is something that is isolated. What you end up with can be multiple isolates that are some or all subsamples of the same organism (e.g. Smith et al. 1992, Nature 356:428-431) or you can get isolates that represent multiple different organisms even from a single plant (e.g. http://oregonstate.edu/dept/coarc/sites/default/files/publication/97_verticillium_isolates_mint.pdf and the above reference from Dr. Platero). In your example, if you select only one isolate per plant, you would end up with nine isolates but would need to do further testing to determine if any were identical.
How many isolates you should collect and what they represent depends on the type of organism you are looking at. I think , each isolate that specially distinguish in its spesific environmental conditions , which Availabile suitable charachter habitat for this organisms , Regardless that isolates are pathogen or promoting .surely means isolate

Dear! How are you! Isolate is physiological variant or races of a pathogen which causes a different reaction on the same host cultivar, one leading to disease the other to resistance. so number of isolates can be determined by 2n where n is the number of host gene pairs.
