Hi, the main two reasons for low yield could be (i) concentration of plant extract, or (ii) the centrifugation process for harvesting nanoparticles is not enough. Try to concentrate the bark extract, basically for providing more reducing agents. You can also try by increasing the incubation time. Please check the following references.

Thanks Jacob, we are comparing chemically and biologically synthesized magnetite NPs. Most methods use plant leaf extract but I am interested in a reproducible methods using stem bark extract with appreciable yield
