I have troubles with C2C12 differentiation these days.

Does the serum type used in seeding affect the cell differentiation. (Shifted form Gibco to Welgene FBS).

But, the differentiation serum remains to be same (2% HS from Sigma).

The media changes are done each other day (48hr). The frequency of media change is shortened while the number of days increase (24hr to 12hr at last days of differentiation).

But, the issue is the cells do not differentiate, just make branched structures around one cell clusters, and combines with similar neighboring structures.

Elongated myotubes are not formed.
