I am trying to culture mouse BM MSCs freshly isolated through magnetic APC positive selection. The seeding density were 0.5*10^5 and 1*10^5 per well (12-well plate) and cell almost died in both conditions. The medium used was α-MEM media+L-glutamine (Thermo Fisher Scientific) supplemented with 20 % fetal bovine serum (FBS; Gibco), 100 units/ml of penicillin and 100µg/ml of streptomycin, 10ng/ml of fibroblast growth factor (FGF)-basic (R&D Systems). I am wondering if the magnet beads binding to the cells affect cell culture.

The cells were collected from mice femurs and tibia, followed by enzyme digestion (collagenase and dispase) and filtration. Would enzyme digestion affect cell culture?
