Recent observations cite chloroquine as an effective agent against the spread of COVID-19 in vitro, but with little success in vivo. Could it be remotely possible to recover mild doses of the virus in individuals who have showed signs of recovery, and utilize reverse engineering methods to generate vaccines from the virus itself? possibly testing on the conjunctiva which is less susceptible to establishment of infection by the virus? Any help please?

One, you are assuming that the people who control the infection is infected with a milder virus. That's probably not true. There is no data to even suggest that those who die have a "stronger" virus. Right now.. it is all on the health of the host and other existing co-morbidities.
You mean knock out virulence genes from the virus? Let's just say this has been tried for a LOT of pathogens...
By far the easiest is to see what worked with SARS and MERS...
