Dear all,
I fixed my gonad tissue with 10% Neutral Buffer Formalin and the nuclear of testicular cells were not clear to see. Is there any way that we can re-fix (by using Bouins) the tissue after sectioning and place on glass slides?
It would be great if could give me some suggestion for this situation. Thank you very much!

Yes. Bouin's is the best fixative for gonadal tissue. We have been using this fixative routinely for the gonadal histology of fish, amphibians and mammals in our lab. But once the tissue is fixed, processed, embedded and sectioned, it may not be possible to go for refixing the same tissue to improve the stainIng intensity. Instead you may try any other nuclear stain to obtain better clarity of nuclear morphology of testicular cells.
