Hi Erman, What do you mean by differential isolation? I routinely isolate neurons, microglia & astrocytes from mouse brains so am able to share my protocols with you if this is what you're looking for? For cortical neurons (do u mean neurons by nerve cells?) I use E13-14 embryos and for glial cells (both microglia & astrocytes) I use P1 mice
Thank you very much for your reply. By differential isolation, I mean that I want to have two different cell population; the first one is nerve cells (brain cells) and the other one is glial cells (microglia). I will reserach the genome wide effects of electromagnetic waves on brain cells. Thus, I would be pleased if you could share your protocols.
Sure - I'm attaching two files here, one for neuronal culture, and another one for microglial culture. Note that as i mentioned earlier, for optimal yields, you'll need to use two different ages of pups to isolate cells : E14 for cortical neurons and P1 for Microglia. Hope they help :)
