Many of my immunoblots look like the ones in the attached photo with that strange form in the middle I tried different things during the handling but they still appear.
I am not sure what you mean with "strange form". The white lines that appear at several points of the stripes are "negative staining" by abundant protein. It´s so tightly packed on the membrane that there can´t develop a background, like writing with a pen on the membrane. The drop like structure in the positive stripes could be a product of air bubbles, high salt concentrations, contamination with genomic DNA...
Hi, could you write somo detail about your protocols? Did you have problem with lysis buffer? Did you have this result with all the incubated antibodies?
