I need quantify the protein amout from human DCs lysed by RIPA buffer ( 150 mM NaCl, 1% NP-40 or Triton X-100, 0.5% sodium deoxycholate, 0.1% SDS, 50 mM Tris, pH 8.0 ) to use in a immunoblot.
Some protein quantification methods are not suitable with some chemicals present in this buffer composition.
Qubit™ Protein Assay Kit is suitable for my purpose?
Qubit doesn't do very well when there are detergents in a solution, and you have several of them. You can still give a try, but I would consider other quantification methods instead of Qubit.
Amanda Rocha Personally, i have been using BCA for quite a time now, sensible and detergents don't really bother the assay, I use the same buffer to do my calibration curve and i think that the results are accurate and reproducible, but you can search on the net for other methods that can be quite good.
