I am doing a intrahepatic immune cell isolation. They ask for 40% optiprep but I currently only have percoll. Could I just use percoll instead ? What is the benefit of using optiprep?
Dear Boram Ham, yes! You can use Percoll for isolation of intrahepatic lymphocytes. Briefly, the method: livers should be perfused with 10 ml PBS before removal, passed through a stainless steel mesh, and suspended in Earle's balanced salt solution. After one washing, resuspend cells in 35% Percoll containing 100 IU/ml heparin, and then centrifuge at 500 X g for 10 min at RT. Resuspend the pellet in RBC lysis solution (0.17 M NH4CI, 0.01 mM EDTA, 0.1 M Tris, pH 7.3), then wash twice in 1% BSA-Earle's balanced salt solution.
