I would like to know the changes in crystallinity of polymer film, caused by biological degradation.
As of my basic knowledge acquired by other literature, either FT-IR or XRD analysis can show the crystallinity changes of polymers.
Could you give me a specific method (required equipment setting, program, material pretreatment, etc.) or recommend me any pieces of literature that I can refer to?

You can consider doing differential scanning colorimetry (DSC) analysis of your polymer films.

In general, polymer crystallinity is usually evaluated using DSC, XRD or pycnometry (from density). Spectroscopy (FTIR, Raman, NMR) can also be used, but that depends on the polymer you are studying - what signals could be used to evaluate the crystallinity.
It would help if you could share what polymer you need to measure and also the conditions of your experiment. You are mentioning biological degradation - the used matrix could complicate the analysis, especially if you are using a complex medium with proteins etc. that could strongly adhere to the polymer.

If your polymer film is stable under x-ray and thickness is more than 200-250 nm, then XRD may help you to study it's crystallinity otherwise HRTEM along with SAED is suitable.
The degree of crystallinity can be determined by several experimental techniques: among the most commonly used are (i) X-ray diffraction, (ii) calorimetry, (iii) density measurements, and (iv) IR. ... If the sample is anisotropic, a suitable average of the diffracted intensity in reciprocal space must be obtained.

Now I am doing a biodegradability test for biodegradable polymers in film shape with 20 - 25 ㎛ thickness that is kinds of PHA (polyhydroxyalkanoates)s or PLA and others. To see the changes caused by microbial activity, I analyzed ATR-FTIR analysis.
The only available methods I can use are ATR-TIR and XRD.
I would like to know any information about defining crystallinity or its changes by those two.
@Could you recommend me some references or give me any information about the films?
