HELA cells are originated from cervical cancer. Virus is very specific for cells. They need particular receptor in the cell, then only then only they can infect.
Anyways the protocol will be the same. We use to follow the same protocol for each infection. Like 70-80% mono layer, then remove the media, and add virus (volume on the basis of flask you are using). Allow it to adhere for 1 hour, and then add media.. keep it for 2-5 days in incubator with 5% of CO2. Observe flask from the second day of inoculation, on 4 or 5 day u will be able to see the plaques. If virus get the receptor then only it will infect the cells, other wise it will not.
It is possible to infect Hela cells by dengue 2 virus, but the infection is not very efficient. Other cell lines are more effective. For example: C6/36 (from Aedes albopictus), A549, HepG2, BHK21, LLC-MK2, Vero ...
