I am planning to do an IHC study from mouse coronal brain sections and will be looking at various tight junction proteins. I of course realize that endothelium is present throughout the brain but I was wondering if anyone was familiar with a region that has particularly high expression of tight junction proteins?
Blood brain barrier properties are primarily determined by junctional complexes between the cerebral endothelial cells. These complexes are comprised of tight and adherens junctions. Therefor you can locate tight junctions nearby blood capillaries in IHC of mouse coronal brain.
Infact In my lab we also looked for same.

In gereneral cortex, either frontal or parietal is the most widely used region for TJ staining in brain capillaries.
The region should also depend on your specific interest and the project. In our current study we stain both frontal cortex and hippocampus. We got great staining in both regions. Be prepared that you need a high magnification confocal image and a longitudinal section of the vessel to get a clear nice picture.

regions of the BBB are best, as they are likely some of the tightest tight junctions in the mammal. As suggested already above, cells of the choroid plexus and capilary vasculature are the ones you want. they would have the highest TEER, and therefore the best tight junctions form you IHC study
