Could anyone know how to troubleshoot the fragment analysis of trisomy? I have noticed that the peak's height are very low (10-20)

We use panel A (D13S258, D13S37, D13S63, D13S796, D18S1002, D18S391) and B marker (D18S851, D18SS535, D21S11, D21S1411, D21S1435, D21S1446). In addition, we do back up marker if marker is missing for reporting.

We found that some peak like D18S851 (from B Marker) is missing in the positive control (male DNA 9948)? Why does it happen? We already increase the injection time to 40 (standard is 24) Should I open another new master mix?
