We have analyzed glutamate decarboxylase autoantibodies (GAD65) in DM serum patients using ELISA and we got concentration (ng/ml). How to determine cut off point of GAD65-Ab because we don't have any literature to calculate cut off points and determine positive and negative from the data.

There are two parameters to consider:
(1) The limit of detection (LOD) or limit of quantitation (LOQ), which relate to the sensitivity of the measurement. Is the measurement sensitive enough to detect autoantibody levels that are present in patients who have the disease?
(2) The differential level between patients and negative controls. What is the baseline level? How much variation is seen in people who don't have the disease?
