I'm working with steroids, so APCI ionization is obvious choice. I have Waters UPLC-MS system with single quad and hybrid ESI/APCI ion source (they call it ESCI, its ESI probe with corona discharge pin so you can do both ESI+APCI). But, my samples do not ionize well. With direct injection I am able to record spectra of pure standard (progesterone for example) but when injected through LC the signal is so weak (super small peak, barely noticeable). I also noticed big fluctuation of current on my corona pin (its set on 5uA, but its always between 3-6) Also the plasma discharge at corona tip is really weak and is sometimes visible but most of the time is invisible. Is this normal? Should I buy new corona pin? I'm somewhat new to APCI technique.
