There is this peculiar problem I am facing in my lab regarding self contamination of the sterilized nutrient broth, the issue is that one specific film forming bacteria is contaminating my sterilized nutrient broth, from my experience it is most probably bacillus subtilis or any other bacillus spp.

Factor 1 - The autoclave is working fine which I checked with autoclave tapes and I don't have enough time to carry out tests with Bacillus stereo. strips which is time consuming.

Factor 2 - I am using Peptone, yeast extract, beef extract and sodium chloride to prepare my nutrient broth rather than using prepared medium (there could be a possibility of any component being contaminated but as I checked before they were clean enough so I'm keeping on hold that possibility for the moment)

Factor 3 - The water I am using is normal drinking water with TDS around 800mg/L and I am really not sure if that is what is causing the problem.

There are other factors too like glassware sterilization, bioburden etc. but I am counting them out because it is just nutrient broth and I am having one specific bacterial contamination. It could be some highly heat resistant spores but in general using an autoclave at 121 degree and 15 psi with volume dependent time is supposed to kill everything.
