Hello!

I have been attempting alkaline Comet Assay with adherant cells. In my 0Gy controls, I have cells with no damage and cells that are ballon shaped (pic) within the same slide. I'm starting to think this is representative of something else than DNA damage?

Secondarily, some of the wells have a lot of background, making it difficult for the analysis software to quantify. We have tried cleaning the slides with ethanol in case it was dust and have tried rinsing samples in case it was cellular debris. Any suggestions as to why I'm seeing so much background?

Ive attached two examples, 1 with a lot and 1 with little background. You can also see the "normal" comets in these to compare to the 0Gy one.
