I noticed that in many gene expression experiments, people combine the gene of interest to eGFP, Does anybody know the advantage of this method ? Is it only for easy detection of the positive clones?

The advantages of tagging the gene of interest to GFP are
1. easy screening and sorting
2. analysis of expression kinetics
3. the subcellular localization of the expressed protein
4. colocalizations with other proteins in the cells
5. tracing the cells during adoptive transfer during animal experimentation etc.
Some of these advantages are useful only in the case of fusion of the gene with GFP. And not all genes are compatible with this kind of fusion.
There are disadvantages also. The fusion of eGFP to a gene of interest could alter functions and sub-cellular localization. Therefore, fusion with smaller tags is preferred for the functional studies.
