what do you want to do after? do you try to centrifugate at 3000g. It's sufficient to eliminate cells and keep viruses

You may consider a panning or rosetting technique in combination with a CMV glycoprotein antibody followed by ficoll banding. Use versine instead of trypsin.

I am sorry I want to keep only CMV infected cell if I do 3000 rpm centrifugation does this may collapse cell. I want to keep those cells to initiate cell to cell infection

To my observation on Day 4 post infection CPE quite visible in the form of Spindle shape cell, these cell are highly virimic and have the capeability to progress cell to cell infection quit rapidly. But the problem is during the seed prepration i have several different level of CMV infected cell those are not good to produce a good infection

You may wish to stain for MHC-I surface expression and make a MACS sort on the MHC(low) population. CMV downregulates MHC-I expression and virus infected cells will have decreased MHC-I levels (uninfected cells, obviously not, as they will not express US3, US6 and so on).

Please consider that your CMV-infected cells become more fragile, bacuse their cytoskeleton is disrupted (hence, they are cytomegalic). This means that sorting in a FACS machine may be too harsh for them to survive, but separation on a MACS column may work well.

more over what kind of filters need to be use eithe negative or positive.

as you said in your suggestion US3 and US6 are membrane markers for labelling Megnatic beeds with if iam not wrong, Please advice?
