stem markers will not tell you anything about clonality
animals; you need a reporter gene such as that in the Brainbow mouse
humans; you need a mutation or single cell cloning if you are talking about cell culture.
Clonality can be tested only by single cells cloning, if you have problem to culture your cells as single cells you can label them and then culture them in single cell density together with other non-labeled supportive cells and to show that your single cell create a a fully labeled clone
If I want to confirm the clonality of stem cells and its daughter cells,I have to prove the daughter cells are originated from a single cell and the genetic content is same in the daughter cells or both?should I consider the extra chromosomal inheritance also?
Both, you need to demonstrate that there is a genetic marker that the stem and daughter cells share, copy number could be one such marker.
