hello, I need your kind advice on how to prepare chitinase detection medium. I have prepared mineral base media fortified with 2% colloidal chitin but yet there is a difficulty in attaining a clear zone around the colonies.

In screening for chitinase using colloidal chitin depends a lot on the colloidal chitin you use, I have been doing it several times, some colloidal chitin doesn't permit seeing the halo until you stain it using congo red afterwards. I used ph indicator (bromo cresol purple) wchich leaves very clear the halo.

if you prepare colloidal chitin from chitin powder (sigma) instead of using chitin flakes you will get prominent zone hopefully. because i did face the same problem but after using powder chitin, somehow the issue solved
