Hello everyone,
recently I have started working with cell-penetrating peptides (CPPs) and I need to produce and purify them. I have a plasmid with a cargo attached to a CPP. In a construct, there is also a signal sequence and a His-tag. My question is, is it possible to produce the peptide in HEK F cell line? I did it once and I didn't get any protein. My concern is that CPP might re-enter the HEK F cells after being synthesized and exported to the medium.
In all the papers I have found, they produce CPPs in E. coli strains. Is there a reason behind it? Thank you.
