I have done cell cycle analysis of sychronized HEK-293 cells incubated with some nanomaterials and in their absence (control) The results show almost no difference in G1 phase, while a decrease in S phase, accompanied by an increase in G2/M phase appears. Could somebody give me some more insight into this data?
* cell cycle analysis was performed three times and these are the mean values. Sample 1 is the lower tested concentration and sample 2 is the higher tested concentration.
In both tested concentrations, no cytotoxicity is observed. On the contrary, the number of cells, when i add the nanomaterials, is increased compared to control.
HEK-293 cells are tranformed and lack proper cell cycle controls that are mediated by Rb and p53 tumor suppressor proteins. Therefore, a moderate increase in cells in the G2/M phase following a treatment of "synchronized" cells is independent of normal cell cycle control and could be attributed to a number of cell signaling pathways activated by the treatment. Consequently, it is harder to interpret the data. One may find a number interpretations in the literature depending upon cell culture conditions.
