I have prepared Hydrogel desks (6 mm) and placed then in 24 well-plate, afterwards, I seeded MSCs cells on top.
there is no cells been attached to the hydrogel desk?????
- cell density was 50000/well.
- I could not observe any cells adhere on the hydrogel desks?

Cells will not adhere to alginate. It does not present any ligands to which mammalian cells can bind. It also does not bind proteins to which cells can binds (e.g. fibronectin, collagen, laminin, etc.). It is possible to graft cell adhesion peptides to alginate to enable attachment. The chemistry is fairly straightforward. I am attaching a couple of papers that document how to make the modifications and the effect is has on cell adhesion.

alginate like hyaluronic acid or any other unmodified polysaccharide, has no binding sites for cell attachment. On the contrary, it has anti-adhesive properties. Unless you chemically modify it or you "attach" binding proteins on its surface, cells will not adhere to it.
You can easily find online protocols for simple modifications that allows cell ashesion.

In addition to the previous answers and to induce the cell attachment, you can either modify the surface of the alginate with certain peptides (e.g., GRGD, RGDS,..), or prepare a hydrogel of alginate with other polymer to which the cells can attach (e.g., chitosan).

As suggested above, I would also recommend you to modify your alginate with RGD adhesion sequences by means of carbodiimide chemistry. It takes just a couple of weeks to complete the whole process. As a pupil of Dr. Mooney's school, we use the following synthetic peptide:
H-Gly-Gly-Gly-Gly-Arg-Gly-Asp-Ser-Pro-OH

I am agree with prof Lawrence Bonassar , the main problem of Alginate materials is that leak of cell adhesion and cell attachment, there many techniques for improving this problem such as using extracellular proteins matrix laminin, fibrnectin collagen…..
