We are currently growing CaCO-2 cells which we are struggling to break the clusters up, is there any tips for this?

Additionally, we are trying to mimic in-vitro so we want to carpet a layer of cells to mimic the intestine, with media above and below it to see where extracellular vesicles will move, but just unsure what is a good amount of cells to plate onto this? We're putting them into 6 well plates and using a mesh scaffold which im not too sure the surface area but I think its about the same as a 12 well plate. Any advice appreciated.
