I want to analyze the net charge of a egg in blastulla stage.

That sounds good. But it is hard to check the net charge of a particular egg. You can check them as a whole by using PBS. Try it.

I tried.. But the question arises what could be the probable explanation of net charge.. whether it is because of chorion bilayer or due to the internal embryonic matter and chorion.

Very good question Suresh. As far as I know the net charge depends on the chorion bilayer and if there is no convincing report published so far then you can do couple of additional tests to confirm your findings and be the first one to report.
