Now I am using PC12 cell line (a Pheochromocytoma cells from rat adrenal medulla) to screen my samples protection against H2O2 toxicity. I want to test one more Neuron cell line. Please help me to choose. Thank you very much in advance.
a lot depends on what you want to find out. if the read-out is cell death and you are working in high-throughput, then you may consider neuro2a cells (need differentiation) , sk-n-mc or sh-5y5y (neuroblastoma cells, grow fast) but they are not so neuronal. Hib5 (principal cells, hippocampus9 are also good, but they require growth and differentiation at different temperature. medullary raphe cell line such as rn33b or motoneuron-like nsc-34 may be an option if you want a specific neuronal type (serotoninergic or cholinergic, respectively) for your exps. good luck....
I will go with Neuro2a because the differentation protocol could be easier than in other cell lines. Also, both Neuro2a and P19 are mice cells, and that could be interesting too since you are working with a rat model.
We use primary mouse cortical neurons from a E15 CD-1 mouse to test for bio-active compounds. It is more work but a prep yields around 100 x 10^6 cells +/- 20% depending on the number of embryos. We also culture them with astrocytes obtained from a P0-P1 mouse.
