We want to analyse Citrate synthase in an endothelial cell line and have a reduced sample volume so we cannot lose it performing tests. Could you suggest a good antibody and its best working dilution? Thanks a lot!
Without knowing your species of interest it may not be appropriate to make any comments/advice.
Here is a suggestion though I never worked with citrate synthase (CS) but from my experiences with other Abcam antibodies tell that these are good.
You may think of trying Abcam Anti-Citrate synthetase rabbit polyclonal antibody (ab96600). This antibody has a wide variety of species selectivity including mouse, rat, human, zebrafish with a predicted cross reactivity with cow and pig.
The data sheet provided reports for validation of this antibody against HepG2, Zebrafish skeletal muscle and mouse liver mitochondrial homogenate in WB. Off course you can have better ideas from researchers in the CS area.
You can email Wen-Tsan Chang (([email protected]) for further information as he is working in this area (http://www.nature.com/srep/2012/121030/srep00785/pdf/srep00785.pdf)
I am sharing the ab96600 data sheet for further reading.
I have worked with the anti-CS from Sigma-Aldrich (C5498) in fibroblast cells and the results are good. I use a 1:2,000 dilution and incubation over night, although I've also obtained good results with a 2-3 hour incubation.
