I am trying to fix protein molecule on carbon grid. I want to take TEM image. i am finding difficulties in fixing protein on carbon grid. is there any modified carbon grid available ?
I think that it is necessary to treat the grids to make them hydrophilic. I would suggest to use pure carbon film on Cu grids (400 or 600 mesh). This will give you lower noise. Then use a glow discharge device to treat the carbon film (made them hydrophilic; grids activation). It is generally recommended to treat the grids with glow discharge immediately before sample preparation.
The glow discharge devices are now quite common in EM labs. A long time ago, we have described a modification of sputter-coater for glow discharge activation of carbon support films (http://doi.wiley.com/10.1002/jemt.1060160304).
